In general, IVRT studies involve a comparison of API release rates between a reference (R) and a test (T) product using six diffusion cells each, in accordance with the USP compendial method. The statistical test is then applied to compare the release rates from eighteen cells for each T and R products.

What is in vitro release?

The in vitro release study is a critical test to assess the safety, efficacy, and quality of nanoparticle-based drug delivery systems, but there is no compendial or regulatory standard. Interestingly, the DM method appeared to misestimate the release kinetics of nanoparticles through separate mechanisms.

How is IVRT measured?

IVRT is measured in apical five-chamber view (A5C), using pulsed Doppler. Sample volume is placed between the aortic valve and the mitral valve, which allows it to record both the closure of the aortic valve and the opening of the mitral valve. IVRT can be measured according to Figure 4 below.

A normal IVRT for a middle-aged adult is approximately 80 msec. A short IVRT (<60 msec) indicates an early mitral valve opening; a long IVRT (>100 msec), a delayed LV relaxation and a late valve opening.

What is normal EE ratio?

In normal individuals the E/e´ ratio is <8. In the presence of diastolic dysfunction / impaired relaxation, e´ will be rather low. In contrast, the E-wave increases with elevated filling pressures. Thus the E/e´ ratio will increase in the presence of diastolic dysfunction.
